چکیده

AIM To identify and characterize the subunit structure of interleukin-13 receptor (IL-13R) in human medulloblastoma cell lines and target them with a chimeric fusion protein consisting of interleukin-13 and Pseudomonas exotoxin (termed as IL-13 cytotoxin). METHODS Five human medulloblastoma cell lines were examined for the expression of IL-13R subunits at the mRNA and protein levels by reverse transcriptase-polymerase chain reaction (RT-PCR) and indirect immunofluorescence studies, respectively. In addition, IL-13 cytotoxin-induced cytotoxicity was examined in these medulloblastoma cell lines by measuring protein synthesis inhibition. RESULTS All five medulloblastoma cell lines expressed mRNA and proteins for IL-4Ra and IL-13Ra1 chains, whereas three of the cell lines also showed the presence of IL-13Ra2. mRNA or protein for IL-2gc chain was not detected in any of the cell lines. Consistent with the expression of IL-13Ra2 chain, IL-13 cytotoxin was highly and specifically cytotoxic to three of five medulloblastoma cell lines. The sensitivity of medulloblastoma cell lines to IL-13 cytotoxin could be completely eliminated by concurrent incubation with excess of IL-13, but not with IL-2 or IL-4. CONCLUSION These studies establish IL-13R, in particular IL-13Ra2, as a medulloblastoma-associated target for IL-13 cytotoxin therapy. IL-13 cytotoxin may be useful for medulloblastoma therapy. Alternatively, IL-13Ra2 may serve as a tumor-specific antigen for active immunotherapy.

Cloning, Expression and Characterization of Recombinant Exotoxin A-Flagellin Fusion Protein as a New Vaccine Candidate against Pseudomonas aeruginosa Infections

Background: Infections due to Pseudomonas aeruginosa are among the leading causes of morbidity and mortality in patients who suffer from impaired immune responses and chronic diseases such as cystic fibrosis. At present, aggressive antibiotic therapy is the only choice for management of P. aeruginosa infections, but emergence of highly resistant strains necessitated the development of novel alt...

Receptor for interleukin 13 on AIDS-associated Kaposi's sarcoma cells serves as a new target for a potent Pseudomonas exotoxin-based chimeric toxin protein.

AIDS-associated Kaposi's sarcoma (AIDS-KS), the most common malignant complication of human immunodeficiency virus infection, is characterized by neoplastic proliferation of mesenchymal cells. AIDS-KS cells release and respond to an array of cytokines through specific plasma membrane receptors. Specific targeting of potent cytotoxic agents to cell surface receptors/antigens on Kaposi's sarcoma ...

Targeting of interleukin-13 receptor on human renal cell carcinoma cells by a recombinant chimeric protein composed of interleukin-13 and a truncated form of Pseudomonas exotoxin A (PE38QQR).

We have previously shown that human renal cell carcinoma (RCC) cells express large numbers of interleukin-13 receptors (IL-13R), a newly described hemopoietic growth factor receptor. To target tumor cells that express IL-13R, we have produced a chimeric protein composed of human IL-13 and a derivative of Pseudomonas exotoxin A, termed PE38QQR. We report here that IL13-PE38QQR is highly cytotoxi...

Human glioma cells overexpress receptors for interleukin 13 and are extremely sensitive to a novel chimeric protein composed of interleukin 13 and pseudomonas exotoxin.

Recently, we have demonstrated that a spectrum of human adenocarcinoma cell lines express binding sites for interleukin 13 (IL-13). These cells are killed by a chimeric protein composed of human (h) IL-13 and a derivative of Pseudomonas exotoxin, PE38QQR (Debinski et al., J. Biol. Chem., 270: 16775-16780, 1995). The cell killing was hIL-13- and hIL-4-specific, indicating that a common binding s...

Targeting interleukin-4 receptors for effective pancreatic cancer therapy.

We demonstrate that pancreatic cancer tissues express receptors for interleukin (IL)-4 in situ at high density. Using the approach of selective receptor targeting, we have tested the efficacy of a recombinant cytotoxin IL4-Pseudomonas exotoxin A, which is composed of a targeting moiety (IL-4) and a mutated form of Pseudomonas exotoxin. Our results demonstrate that this molecule exerts vigorous ...
